Effect of Antiinflammatory Drugs on the Degradantion of Hyaluronic Acid by Fe^++, H_2O_2 and Ascorbate

Abstract
Hydroxyl radicals(OH¡¤)are produced when H_2O_2 react with O?? or ascorbate in the presence of metal ions, particularly iron(11). It has been proposed that some antiinflammatory drugs may exert their therapeutic effects by inhibiting production of reactive oxygen products on tissue components. Thus, in the present study effects of reactive oxygen products generated by Fe^++, H_2O_2 and /or ascorbate on viscosity of hyaluronic acid which maintain lubricating properties of synovial fluid were examined. Effect of some antiinflammatory drugs was observed on reduction of viscosity of hyaluronic acid by Fe^++, H_2O_2 and ascorbate. 1. When hyaluronic acid was incubated with Fe^++ and H_2O_2, its viscosity was reduced with time and their effects was markedly enhanced by ascorbate. At the concentrations used, Fe^++, H_2O_2 or ascorbate showed little or no effect on the viscosity. 2. Reduction of viscosity of hyaluronic acid and ethylene production from methional by Fe^++, H_2O_2 and ascorbate were effectively inhibited by catalase or OH¡¤ scavenger, mannitol, sodium benzoate and sodium formate. There results suggested that the reduction of viscosity of hyaluronic acid was due to OH¡¤. 3. The reduction of viscosity of hyaluronic acid ethylene production by Fe^++, H_2O_2 and ascorbate was effectively inhibited sodium salicylate, acetyl salicylic acid, imidazole, indomethacin and hydrocortisone. Their effects were increased at higher concentrations. The results obtained suggest that antiinflammatory effects of some antiinflammatory drugs may be attributed to their quenching effect on the hydroxyl radicals(OH¡¤) which are considered to be the most reactive and the most likely immediate oxidant of the destructive processes.
